자동화에 중점을 두고 개발

 

Cloud Alpha MOVE는 Corning ® HTS Transwell ® 96-웰 투과 지원 시스템과 12- 및 24-웰 크기의 삽입물에서 포유류 세포 배양을 노출할 수 있습니다. 클라우드 시스템은 용액 및 현탁액의 분무화에 적합합니다. 응용 분야는 흡입 약물의 스크리닝, 화학 물질 또는 나노 입자와 같은 흡입 물질의 독성 테스트 및 바이러스 연구입니다.

 

세 가지 유형의 분무기 선택

 

액적 MMAD 범위가 2.5–6.0 µm, 2.5–4.0 µm, 4.0–6.0 µm인 3가지 유형의 진동 메쉬 Nebulizer와 9.0–12.0 µm의 특수 버전이 함께 제공됩니다. 이 장치는 소량의 테스트 재료를 사용할 수 있을 때마다 테스트에 특히 적합합니다.

Air/Liquid Cultivation

In response to the scientific need to expose in physiologically relevant conditions, the VITROCELL® Cloud Alpha exposure devices have been specifically designed to enable direct exposure of mammalian cells or tissue at the Air/Liquid Interface. Here the cell cultures are not covered with media as opposed to submerged conditions which cause an undesired inter­action of the formerly airborne substances with the culture media.

Cell systems cultivated on membrane inserts are exposed at the Air/Liquid Interface (ALI) so that the test substances are received directly. This approach allows for more credible and authentic results than by submerged exposure due to a closer replication of the human physiology.

For each of the exposures applies: After each nebulization the exposure chamber is automatically evacuated by vacuum pump to enable equal exposure conditions for the following nebulisation.

VITROCELL® exposure systems have been specifically designed and engineered to give researchers the possibility of directly exposing mammalian cells or tissue at the air/liquid interface. Thus, all cell types cultivated on microporous membranes can be used. This approach allows for more credible and authentic results than by submerged exposure due to a closer replication of the human physiology.

The Quartz Crystal Microbalance sensor is integrated in the Cloud Alpha MOVE exposure module. It is capable of measuring the deposited mass in nanogram/cm2. Results are recorded in the VITROCELL® Monitor software. Data is presented in graphs and stored in MS Excel®.

Ideal for small quantities of test substance

The VITROCELL® Cloud Alpha MAX is designed for lower nebulization volumes and very high deposition efficiency. This is important when only small quantities of material are available or when expensive test substances need to be tested. The non-cell exposed area is reduced significantly by direct nebulization into exposure chambers with reduced chamber volume. A set of 3 nebulizers allows for simultaneous testing of 3 concentrations, or a combination of Microbalance, control liquid and suspension of the test substance.

After pipetting the suspension, exposure to the cell cultures takes place in three phases:

Choice of three types of nebulizers

The system comes with a choice of 3 types of vibrating mesh nebulizers having droplet MMAD ranges of 2.5 – 6.0 µm, 2.5 – 4.0 µm and 4.0 – 6.0 µm. Recommended nebulisation volumes are 10 – 40 µl. So the device is particularly suitable for testing whenever small quantities of testing materials are available.

The QCM sensor is integrated in the Cloud Alpha MAX exposure module. It is capable of measuring the deposited mass at a resolution of 10 nanogram/cm2 per second. Results are reported online by the VITROCELL Monitor Software. Data is presented in graphs and stored in MS Excel®.
